Virus Sampling Tubes for Nucleic Acid Testing In the realm of nucleic acid testing, more specifically polymerase chain reaction (PCR) fluorescence methods, the process begins with sample collection—a task that is crucial to its accuracy. Among the various components involved,virus sampling tubes play an instrumental role. The uninterrupted supply of these tubes is vital for effective disease prevention and control measures. But what exactly are virus sampling tubes, and what are their constitutive reagent components? Let’s delve into that:
What are Virus Collection Tubes and Their Functions? Virus sampling tubes are employed by health regulatory and clinical institutions for the collection and transportation of infectious microorganisms. They are ideal for collecting and transporting a variety of viruses including Influenza viruses (such as common influenza, highly pathogenic avian influenza, and Influenza A H1N1), Hand, Foot and Mouth Disease virus, and other pathogens, including newly emerging coronaviruses. Furthermore, these tubes are useful for certain types of bacteria, such as mycoplasma, chlamydia, and ureaplasma, and are designed for short-term storage at temperatures between 2-8℃ and long-term preservation in a -80℃ refrigerator or liquid nitrogen.
Typical samples include throat swabs, nasal swabs, sputum, bronchial lavage fluid, and alveolar lavage fluid. These virus sampling tubes, which are RNase-free to safeguard RNA integrity, are used for collecting samples from the site of infection through to the laboratory testing process. Once collected, laboratory technicians transport the nasopharyngeal swabs or tissue specimens to undergo PCR extraction and further testing.
What are the Composition and Main Components of a Virus Collection Tube?
A virus sampling tube consists of three components: the swab, the sampling tube, and the virus preservation solution.
Virus preservation solutions include Hank’s solution base, gentamicin, fungal antibiotics, BSA (V), cryoprotectants, biological buffers, and amino acids. Based on Hank’s solution, additional virus stabilizing ingredients like 4-hydroxyethylpiperazineethanesulfonic acid (HEPES) and BSA (bovine serum albumin fifth component) are incorporated to maintain the activity of the virus over a wide temperature range. This combination slows down viral decomposition and increases the likelihood of successful virus isolation.
Another preservation solution is prepared using a nucleic acid extraction lysis solution as the base. The primary ingredients include balanced salts, EDTA chelating agent, guanidine salts (like guanidine isothiocyanate and guanidine hydrochloride), anionic surfactants (such as dodecane sodium sulfate), cationic surfactants (like tetradecyltrimethylammonium oxalate), phenol, 8-hydroxyquinoline, dithiothreitol (DTT), proteinase K, and other components. This preservation solution directly disrupts the virus to release nucleic acid and neutralizes nucleolytic enzymes (like RNase). It is particularly suitable for RT-PCR. However, it can inactivate the virus, rendering it unsuitable for viral culture and isolation.
It is recommended to use EDTA salts (like dipotassium ethylenediaminetetraacetate and disodium ethylenediaminetetraacetate) as chelating agents in virus preservation solutions. It is advised against using heparins (like sodium heparin and lithium heparin) to avoid adverse effects during PCR testing.
The quality of the virus preservation solution is crucial for enhancing the positive rate of PCR detection and successful virus isolation. It ultimately determines the quality of the virus sampling tubes. Incorporating room-temperature stable fungal antibiotics can address mold contamination issues post-sampling and enable room-temperature storage of the sampled liquid.
